RESUMEN

The cotton aphid Aphis gossypii Glover is an important cotton pest, and means of controlling this insect is a primary research focus. Although biological rhythm is an important mechanism that regulates numerous insect processes and activities, its role in cotton aphid has not been elucidated. In the present study, four highly-expressed circadian rhythm genes were selected from the cotton aphid genome database and their physicochemical properties and protein structures were analyzed. These genes were in the Takeout, Timeless, and Timeless interacting-related families, and the corresponding proteins contained highly-conserved Swis and TIMELESS domains. Gene expression analysis at multiple developmental stages revealed differing expression patterns between the four genes. AgCLK-1 had the highest relative expression of the four, especially during the nymph period. Silencing AgCLK-1 caused a significant refusal of the cotton aphids to feed at 1, 3, and 5 d of treatment. These results demonstrated that AgCLK-1 played a key role in regulating the feeding behavior of cotton aphid. This new functional understanding provides novel insights into cotton aphid biology and suggests new targeting strategies for agricultural pest control.

RESUMEN

Background: Immunoglobulin A nephropathy (IgAN) and idiopathic membranous nephropathy (IMN) are the most common glomerular diseases. Immunofluorescence (IF) tests of renal tissues are crucial for the diagnosis. We developed a multiple convolutional neural network (CNN)-facilitated diagnostic program to assist the IF diagnosis of IgAN and IMN. Methods: The diagnostic program consisted of four parts: a CNN trained as a glomeruli detection module, an IF intensity comparator, dual-CNN (D-CNN) trained as a deposition appearance and location classifier and a post-processing module. A total of 1573 glomerular IF images from 1009 patients with glomerular diseases were used for the training and validation of the diagnostic program. A total of 1610 images of 426 patients from different hospitals were used as test datasets. The performance of the diagnostic program was compared with nephropathologists. Results: In >90% of the tested images, the glomerulus location module achieved an intersection over union >0.8. The accuracy of the D-CNN in recognizing irregular granular mesangial deposition and fine granular deposition along the glomerular basement membrane was 96.1% and 93.3%, respectively. As for the diagnostic program, the accuracy, sensitivity and specificity of diagnosing suspected IgAN were 97.6%, 94.4% and 96.0%, respectively. The accuracy, sensitivity and specificity of diagnosing suspected IMN were 91.7%, 88.9% and 95.8%, respectively. The corresponding areas under the curve (AUCs) were 0.983 and 0.935. When tested with images from the outside hospital, the diagnostic program showed stable performance. The AUCs for diagnosing suspected IgAN and IMN were 0.972 and 0.948, respectively. Compared with inexperienced nephropathologists, the program showed better performance. Conclusion: The proposed diagnostic program could assist the IF diagnosis of IgAN and IMN.

Cytochrome P450 monooxygenases play important roles in insect metabolism and detoxification of toxic plant substances. However, the function of CYP6 family genes in degrading plant toxicants in Aphis gossypii has yet to be elucidated. In this study, AgoCYP6CY19, an A. gossypii CYP gene that differentially expresses in cotton- and cucumber-specialized aphids, was characterized. Spatiotemporal expression profiling revealed that AgoCYP6CY19 expression was higher in second instar nymph and 7 day old adults than in other developmental stages. Although the expression of AgoCYP6CY19 was significantly higher in cotton-specialized aphids, AgoCYP6CY19 silencing significantly increased larval and adult mortality and reduced total fecundity in both cotton- and cucumber-specialized aphids. What is more, the expression of AgoCYP6CY19 was significantly induced after the cotton-specialized and cucumber-specialized aphids fed on epigallocatechin gallate (EGCG) and cucurbitacin B (CucB), respectively. These findings demonstrate that AgoCYP6CY19 plays a pivotal role in toxic plant substance detoxification and metabolism. Functional knowledge about plant toxicity tolerance genes in this major pest can provide new insights into insect detoxification of toxic plant substances and insecticides and offer new targets for agricultural pest control strategies.

The recent outbreak of COVID-19 has infected a large number of patients, increasing the importance of adequate disinfection of the hospital environment. We conducted this study to explore environmental virus contamination and the effect of terminal disinfection in the isolation ward of patients with COVID-19. A swab kit was used to sample various surfaces in the isolation and observation wards using the smear method. The samples were immediately sent to the PCR department of the laboratory for nucleic acid detection of COVID-19. We analyzed 31 high-frequency contact sites in three isolation wards of actively sick patients, of which seven were positive (22.58%, 7/31). Positive sites included the transfer window, bed rail, buffer room door handle, toilet door handle, and toilet faucet. All 55 samples taken from the wards of cured patients and the wards after terminal disinfection were negative. Virus contamination in areas frequently touched by patients in the isolation ward was high, so the awareness of correct disinfection must be increased. Use of 1,000-2,000 mg/L chlorine-containing disinfectant in the isolation ward was effective.

Given the ever-increasing commercial planting of transgenic plants across the world, an evaluation of their impacts on non-target organisms is as an important part of the risk assessment process. Propylea japonica is a dominant non-target predator and pollen feeder insect that is prevalent in Bt cotton fields, and it is thus in direct contact with Bt proteins. However, the effect of Bt proteins on P. japonica has not received much attention. In this study, the effects of Cry1Ac and/or Cry2Ab proteins on P. japonica were investigated from three aspects. First, no significant differences in the diversity of the microbiota nor change in species composition and community structure were observed among Cry protein treatments. Firmicutes are the most abundant bacterial phylum present in P. japonica, followed by Proteobacteria and Actinobacteria. The most abundant genus was Staphylococcus. Second, the expression levels of the detoxification and digestion-related genes did not change significantly in any Cry protein treatment. Third, none of the Cry proteins affected the population fitness of P. japonica. These results indicated that P. japonica was not sensitive to Bt proteins, suggesting that growing Bt cotton expressing Cry1Ac and/or Cry2Ab will pose negligible risks to P. japonica.
